The specially designed kit includes NAC Buffer for the standard pretreatment of 200 μl – 5 ml sputum samples before lysis of the material. The resulting bacterial cell pellet is dissolved in a special Resuspension Buffer and completely transferred to the Extraction Tube. Optimized buffer and enzymes lyse the samples under a special temperature profile, stabilize nucleic acids and selectively enhance DNA adsorption to the membrane. The membranes are efficiently washed before the nucleic acids are eluted in order to remove all PCR inhibitors and proteins. Based on patented technology, the kit uses buffers without any hazardous chaotropic components. The purified high quality DNA is ready to use for subsequent downstream applications or can be stored at –20 °C for subsequent use:

Different numbers of Mycobacteria genomes (106, 105, 103 and 10 respectively) were mixed with 200 µl lavage fluid. The bacteria genomes were isolated with the RTP Mycobacteria Kit and eluted with 80 µl Elution Buffer. 5 µl were amplified in real-time fluorescence PCR using an ABI PISM 7000 Sequence Detection System.
